DSIP (Delta Sleep-Inducing Peptide) was first isolated from rabbit brain in 1977. It is a nine-amino acid peptide naturally synthesised in the hypothalamus that promotes a specific type of deep sleep characterised by increased delta-wave EEG activity. This delta sleep phase is associated with physical recovery, growth hormone release, and memory consolidation.
DSIP interacts with the hypothalamic-pituitary-adrenal (HPA) axis, modulating cortisol and ACTH secretion. Brain and plasma DSIP concentrations follow a marked circadian rhythm, with low levels in the morning and higher levels in the afternoon. Published research has also demonstrated anti-tumour properties: lifetime DSIP administration decreased spontaneous tumour incidence 2.6-fold in mice.
