MK-677 is a non-peptidic agonist of the ghrelin receptor (GHS-R1a). Unlike injectable growth hormone secretagogues, it is orally bioavailable and produces sustained elevations in circulating GH and IGF-1 without significantly affecting cortisol secretion.
In a randomised, double-blind, placebo-controlled trial involving healthy older adults, 25 mg/day for 12 months restored GH and IGF-1 profiles to levels characteristic of younger adults without significant adverse effects. Additional clinical data has explored its effects on body composition, bone mineral density, and sleep architecture in both young and elderly subjects. MK-677 significantly increased REM sleep duration and sleep quality in published clinical studies.
