Ipamorelin is a synthetic pentapeptide that acts as a selective agonist of the ghrelin/growth hormone secretagogue receptor (GHS-R). What makes Ipamorelin unique among GH secretagogues is its selectivity: it stimulates growth hormone release without significantly increasing cortisol, prolactin, or ACTH levels. Older-generation GH secretagogues like GHRP-6 and GHRP-2 produce broader hormonal responses including appetite stimulation and cortisol elevation.
In preclinical studies, Ipamorelin induced longitudinal bone growth in rats and counteracted glucocorticoid-induced decreases in bone formation. It is frequently studied in combination with CJC-1295 (with or without DAC) to create a synergistic GH pulse: CJC-1295 amplifies the pulse while Ipamorelin triggers it.
